FDA finalizes biosimilar manufacturing-change guidance: an inspection-readiness review
FDA update · Source reviewed October 29, 2026.
FDA issued final guidance in October 2026 on postapproval manufacturing changes to biosimilar and interchangeable biosimilar products licensed under the Public Health Service Act. It finalizes the July 2024 draft. This is a product-specific guidance document; it should not be treated as a universal framework for every biologic or drug.
Our inspection-readiness questions
For an internal review, start with one actual change and map the decision trail. What changed, which product and site are affected, what evidence supports the assessment, and who owns the decision? Keep the regulatory submission assessment connected to the quality-system record.
- Identify the approved baseline and the proposed or implemented state.
- Connect the assessment to supporting manufacturing, analytical, and comparability evidence appropriate to the change.
- Document unresolved questions and the reason for the selected regulatory pathway.
- Check that procedures, training, implementation records, and follow-up activities tell the same story.
These are our suggested review questions, not a restatement of FDA recommendations or a determination of the reporting category for your change. Read the full guidance and applicable requirements before making that decision.
